If you do go the Unity route consider getting 2D Toolkit which will get you a full and proper 2D system with tile maps, multi res assets, a solid animation editor, etc. Id wager a guess and say that with Python and C experience they are gonna slow you down considerably. Once you know how it works then evaluate them. If so, avoid Playmaker and visual scripting tools like the plague until you actually learn how Unity works. You said you've got some C and Python experience. Lots of folks are telling you to use Playmaker. Animations are tedious to deal with (you're stuck in their dope sheet with ridiculously small sprite thumbnails), you're more or less forced into using Mecanim even for a simple two sprite animation, no tilemap support, no multi res support, etc. Be very aware of what Unitys 2D tools consist of (and it's not much). The amount of extra work required to handle multi res assets is nightmarish.
